Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Specificity/Sensitivity

ATP1B3 (F4Z2D) Rabbit Monoclonal Antibody recognizes endogenous levels of total ATP1B3 protein.

Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Cys191 of human ATP1B3 protein.

Background

The sodium/potassium-transporting ATPase (Na+/K+-ATPase) is a transmembrane protein that actively pumps sodium ions out of the cell and potassium ions into the cell, establishing electrochemical gradients that are crucial for various cellular functions. The beta subunits are essential for the proper assembly, trafficking, and stability of the Na+/K+-ATPase, and they also modulate the activity of the alpha subunits (1-3). ATP1B3 is the beta-3 subunit of the Na+/K+-ATPase. ATP1B3, as part of the Na+/K+-ATPase, maintains the electrochemical gradients necessary for nerve impulse transmission that facilitates downstream muscle contraction, nutrient uptake, and cell volume regulation (1-3). The Na+/K+-ATPase is also involved in cell signaling pathways, including those regulating motility and adhesion in normal cells and cancer cells (4-6).

Altered expression of ATP1B3 has been observed in various cancers, suggesting a potential role in tumor progression. It may represent a novel prognostic biomarker in some tumor types, particularly hepatocellular carcinoma, but potentially other tumor types (4-8).

Species Reactivity

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
