Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Specificity/Sensitivity

alpha-2-Macroglobulin (E6N7X) Rabbit Monoclonal Antibody recognizes endogenous levels of total α-2-Macroglobulin protein. Non-specific staining was observed in a subset of kidney tubules by immunohistochemistry.

Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Ala556 of human α-2-Macroglobulin protein.

Background

α-2-Macroglobulin (A2M) is a conserved tetrameric pan protease inhibitor found in many biological fluids, including plasma and cerebral spinal fluid. A2M can bind to most proteases, as well as growth factors, hormones, and cytokines. Clearance of A2M-bound proteins is mediated by low-density lipoprotein receptor-related protein 1 (LRP1/CD91) (1). Blood A2M levels decrease with age in humans, and this decrease may promote tumor development (2,3). In rodents, A2M modulates tumor cell adhesion, migration, and growth through inhibition of PI3K/Akt and SMAD pathways (3). Conversely, interaction with MMP2 may inhibit MMP2 activity in aged rat kidney, resulting in age-related accumulation of ECM and fibrosis (4). A2M may be a useful biomarker for liver fibrosis (5).

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
