Storage

Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Lys252 of human dCK protein.

Background

Deoxycytidine kinase (dCK) catalyzes the rate-limiting step in the nucleoside salvage pathway. dCK phosphorylates salvaged deoxycytidine (dC) to generate deoxycytidine monophosphate (dCMP), a precursor for both deoxycytidine triphosphate (dCTP) and deoxythymidine triphosphate (dTTP) pools (1). dCK also phosphorylates deoxyadenosine (dA) and deoxyguanosine (dG) to generate dA monophosphate and dG monophosphate, respectively. Gemcitabine is an anticancer nucleoside that is a dC analog. It is a pro-drug and must be phosphorylated by dCK to an active form. dCK is phosphorylated on Ser74 by the DNA damage signaling kinases ATM and ATR, and this phosphorylation increases dCK activity (2,3).

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v nonfat dry milk, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
