Background
hnRNP U, also known as scaffold attachment factor A (SAF-A) or SP120, was first described as a nuclear matrix protein that contributes to RNA metabolism (1-3). hnRNP U has been shown to bind pre-mRNAs and contributes to splicing by controlling U2 snRNP maturation (4). Furthermore, hnRNP U has been shown to bind chromatin to help regulate its structure during interphase (5). During prophase, hnRNP U becomes phosphorylated by Aurora-B, which evicts hnRNP U and the chromatin-associated RNAs bound to it, a process critical to proper chromosome alignment (6,7). hnRNP U plays a role in the chromatin localization of Xist RNA, and loss of hnRNP U in ES cells fail to achieve an inactivated X chromosome (8). hnRNP U has also been shown to play a key role in immunity, functioning as a viral dsRNA sensor. Upon viral recognition, hnRNP U oligomerizes and activates numerous antiviral genes, including IFNB1 (9). Loss of function variants of hnRNP U have been shown to cause early onset epilepsy and intellectual disorders (10,11).
