Specificity/Sensitivity
NEK7 (E4E1B) Rabbit mAb recognizes endogenous levels of total NEK7 protein.
Source / Purification
Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human NEK7 protein.
Background
NEK7 (NIMA-related protein kinase 7), is one of a family of serine/threonine kinases, named NEK1–NEK11, which are involved in the control of mitosis and other non-mitotic functions (1-6). NEK7 is the smallest NIMA-related kinase in mammals, and is widely expressed by many tissues (1-3). Its role in mitosis relates to centrosome enrichment and microtubule nucleation, and similar to other NIMA-related kinases, NEK7 is localized to the microtubule-organizing center (MTOC) at the time of mitotic entry (3,6). NEK7 has also been demonstrated to be essential for NLRP3 inflammasome activation by directly binding to the leucine-rich repeat (LRR) domain of NLRP3 (4-6).
Species Reactivity
Species reactivity is determined by testing in at least one approved application (e.g., western blot).
Western Blot Buffer
IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.

NEK7 (E4E1B) Rabbit mAb
Western blot analysis of extracts from various cell lines using NEK7 (E4E1B) Rabbit mAb (upper) or GAPDH (D16H11) XP® Rabbit mAb #5174 (lower). Low expression of NEK7 protein in Hep G2 and mES cells is consistent with the predicted expression pattern.
Western blot analysis of extracts from 293T cells, mock transfected (-) or transfected with constructs expressing Myc/DDK-tagged full-length human NEK7 protein (hNEK7-Myc/DDK; +) or Myc/DDK-tagged full-length mouse NEK7 protein (mNEK7-Myc/DDK; +), using NEK7 (E4E1B) Rabbit mAb (upper), Myc-Tag (71D10) Rabbit mAb #2278 (middle), or GAPDH (D16H11) XP® Rabbit mAb #5174 (lower).
Simple Western™ analysis of lysates (1.0 mg/mL) from Jurkat cells using NEK7 (E4E1B) Rabbit mAb #87795. The virtual lane view (left) shows the target band (as indicated) at 1:10 and 1:50 dilutions of primary antibody. The corresponding electropherogram view (right) plots chemiluminescence by molecular weight along the capillary at 1:10 (blue line) and 1:50 (green line) dilutions of primary antibody. This experiment was performed under reducing conditions on the Jess™ Simple Western instrument from ProteinSimple, a BioTechne brand, using the 12-230 kDa separation module.

NEK7 (E4E1B) Rabbit mAb
Immunoprecipitation of NEK7 protein from Jurkat cell extracts. Lane 1 is 10% input, lane 2 is Rabbit (DA1E) mAb IgG XP® Isotype Control #3900, and lane 3 is NEK7 (E4E1B) Rabbit mAb. Western blot analysis was performed using NEK7 (E4E1B) Rabbit mAb. Mouse Anti-rabbit IgG (Conformation Specific) (L27A9) mAb (HRP Conjugate) #5127 was used as a secondary antibody.
