Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

Specificity / Sensitivity

Siglec-7 (F4O1S) Rabbit mAb recognizes endogenous levels of total Siglec-7 protein. This antibody detects an 18 kDa band of unknown origin in some cell lines.

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Met395 of human Siglec-7 protein.

Background

Siglec-7 (Sialic acid-binding Ig-like lectin 7, AIRM1) is a member of the CD33-related Siglec family, a subset of the immunoglobulin superfamily of cell surface proteins (1-3). Siglec-7 is found in human immune cells such as natural killer (NK) cells, monocytes, and neutrophils (1,2). Siglec-7 acts as an immune inhibitory receptor via its intracellular Tyrosine-based Inhibitory Motif (ITIM) (1-5). Downregulation of cell surface Siglec-7 is an early marker of NK cell dysfunction, which can be associated with chronic viral infection or impaired anti-tumor activity (4).

Western Blot Buffer

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.
