Src Antibody detects endogenous levels of Src proteins. This antibody cross-reacts with over-expressed levels of Yes, Fyn, and Fgr but does not cross-react with other Src family members.

Polyclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to residues close to the carboxy-terminus of human Src. Antibodies are purified by protein A and peptide affinity chromatography.
Background
The Src family of protein tyrosine kinases, which includes Src, Lyn, Fyn, Yes, Lck, Blk, and Hck, are important in the regulation of growth and differentiation of eukaryotic cells (1). Src activity is regulated by tyrosine phosphorylation at two sites, but with opposing effects. While phosphorylation at Tyr416 in the activation loop of the kinase domain upregulates enzyme activity, phosphorylation at Tyr527 in the carboxy-terminal tail by Csk renders the enzyme less active (2).

Species reactivity is determined by testing in at least one approved application (e.g., western blot).

IMPORTANT: For western blots, incubate membrane with diluted primary antibody in 5% w/v BSA, 1X TBS, 0.1% Tween® 20 at 4°C with gentle shaking, overnight.

Src Antibody
Western blot analysis of extracts from various cell lines using Src Antibody.
Immunoprecipitation of Src from A-431 cell extracts. Lane 1 is 10% input, lane 2 is Normal Rabbit IgG #2729, and lane 3 is Src Antibody. Western blot analysis was performed using Src (L4A1) Mouse mAb #2110. Anti-mouse IgG, HRP-linked Antibody #7076 was used as the secondary antibody.
