    Product Description

    The FastScan™ Phospho-Jak2 (Tyr1007/1008) ELISA Kit is a sandwich enzyme-linked immunosorbent assay (ELISA) that detects endogenous levels of Jak2 when phosphorylated at Tyr1007/1008. To perform the assay, sample is incubated with a capture antibody conjugated with a proprietary tag and a second detection antibody linked to HRP, forming a sandwich with phospho-Jak2 (Tyr1007/1008) in solution. This entire complex is immobilized to the plate via an anti-tag antibody. The wells are then washed to remove unbound material. TMB is then added. The magnitude of observed signal is proportional to the quantity of phospho-Jak2 (Tyr1007/1008).

    IMPORTANT: This FastScan™ ELISA Kit requires 4 washes at Step 6 of the protocol.

    Specificity / Sensitivity

    The FastScan™ Phospho-Jak2 (Tyr1007/1008) ELISA Kit detects endogenous levels of Jak2 when phosphorylated at Tyr1007/1008, as shown in Figure 1. This kit detects proteins from the indicated species, as determined through in-house testing, but may also detect homologous proteins from other species.

    Background

    Members of the Janus family of tyrosine kinases (Jak1, Jak2, Jak3, and Tyk2) are activated by ligands binding to a number of associated cytokine receptors (1). Upon cytokine receptor activation, Jak proteins become autophosphorylated and phosphorylate their associated receptors to provide multiple binding sites for signaling proteins. These associated signaling proteins, such as Stats (2), Shc (3), insulin receptor substrates (4), and focal adhesion kinase (FAK) (5), typically contain SH2 or other phospho-tyrosine-binding domains.
