    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human GCSAM/HGAL protein.

    Background

    Germinal center-associated signaling and motility protein/Human germinal center-associated lymphoma (GCSAM/HGAL) is reported to be a marker for germinal center B cells and germinal center-derived B-cell lymphomas (1). Research studies have shown that GCSAM is an adaptor protein anchored to the plasma membrane via myristoyl and palmitoyl linkages and is phosphorylated on multiple tyrosine residues by Syk and Lyn upon engagement of the B cell antigen receptor (BCR), which promotes its interaction with Grb2 and Syk. Binding of GCSAM to Syk positively regulates BCR signaling by enhancing Syk kinase activity (2-5). In addition to regulating antigen-induced BCR signaling in germinal center B cells, research studies have demonstrated that GCSAM restricts motility of B cells within the germinal center microenvironment through activation of RhoA signaling (6). Through regulation of these two processes, GCSAM has been implicated in the differentiation of germinal center B cells to either plasma cells or memory B cells (7).
