    Product Usage Information

For optimal ChIP results, use 5 μL of antibody and 10 μg of chromatin (approximately 4 × 106 cells) per IP. This antibody has been validated using SimpleChIP® Enzymatic Chromatin IP Kits.
    Application Dilution
    Western Blotting 1:1000
    Chromatin IP 1:100

    Storage

Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at –20°C. Do not aliquot the antibody.

    Protocol

    Specificity / Sensitivity

HNF4γ (E4V2B) Rabbit mAb recognizes endogenous levels of total HNF4γ protein.

    Species Reactivity:

    Human, Rat, Monkey

    Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Val363 of human HNF4γ protein.

    Background

Hepatocyte nuclear factor 4γ (HNF4γ) is a transcription factor that belongs to the orphan nuclear receptor superfamily (1). Studies of Hnf4g knockout mice show that HNF4γ is involved in the regulation of glucose homeostasis (2,3). In addition, HNF4γ expression was shown to be upregulated in lung cancer. Overexpressed HNF4γ stimulates lung cancer cell proliferation (1). Furthermore, an aberrant gastrointestinal-lineage transcriptome was identified in about 30% of castration-resistant prostate cancers. HNF4γ, along with HNF1α, regulates this transcriptome. HNF4γ generates and maintains enhancers at the gastrointestinal-lineage genes as a pioneer factor and mediates castration resistance (4).
