    Product Information

    Product Description

This Cell Signaling Technology antibody is conjugated to Alexa Fluor® 647 fluorescent dye under optimal conditions and tested in-house for direct flow cytometric analysis in human cells. This antibody conjugate is expected to exhibit the same species cross-reactivity as the unconjugated Bax (E4U1V) Rabbit mAb #41162.

    Product Usage Information

    Application Dilution
    Flow Cytometry (Fixed/Permeabilized) 1:50

    Storage

Supplied in PBS (pH 7.2), less than 0.1% sodium azide, and 2 mg/mL BSA. Store at 4°C. Do not aliquot the antibody. Protect from light. Do not freeze.

    Protocol

    Specificity / Sensitivity

Bax (E4U1V) Rabbit mAb (Alexa Fluor® 647 Conjugate) recognizes endogenous levels of total Bax protein.

    Species Reactivity:

    Human

    Source / Purification

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Ala42 of human Bax protein.

    Background

Bax is a key component for cellular induced apoptosis through mitochondrial stress (1). Upon apoptotic stimulation, Bax forms oligomers and translocates from the cytosol to the mitochondrial membrane (2). Through interactions with pore proteins on the mitochondrial membrane, Bax increases the membrane's permeability, which leads to the release of cytochrome c from mitochondria, activation of caspase-9, and initiation of the caspase activation pathway for apoptosis (3,4).
