    Product Information

    Product Description

The rapid protocol (RP) PathScan® RP Androgen Receptor (Carboxy-terminal Antigen) Sandwich ELISA Kit is a solid phase sandwich enzyme-linked immunosorbent assay (ELISA) that detects endogenous levels of androgen receptor protein in a reduced assay time of 1.5 hours. Incubation of cell lysates and detection antibody on the coated microwell plate forms a sandwich with androgen receptor protein in a single step. The plate is then extensively washed and TMB reagent is added for signal development. The magnitude of absorbance for the developed color is proportional to the quantity of androgen receptor protein. *Antibodies in this kit are custom formulations specific to kit.

    Protocol

    Specificity / Sensitivity

The PathScan® RP Androgen Receptor (Carboxy-terminal Antigen) Sandwich ELISA Kit detects endogenous levels of androgen receptor protein. The kit sensitivity is shown in Figure 2. This kit detects proteins from the indicated species, as determined through in-house testing, but may also detect homologous proteins from other species. One ELISA antibody recognizes residues near the amino-terminal region of androgen receptor, while the other antibody recognizes the carboxy-terminal antigen that is present in full-length androgen receptor but not in the androgen receptor (V7 isoform) protein.

    Background

Androgen receptor (AR), a zinc finger transcription factor belonging to the nuclear receptor superfamily, is activated by phosphorylation and dimerization upon ligand binding (1). This promotes nuclear localization and binding of AR to androgen response elements in androgen target genes. Research studies have shown that AR plays a crucial role in several stages of male development and the progression of prostate cancer (2,3).
