    Product Information

    Product Description

The rapid protocol (RP) PathScan® RP PIP4K2C Sandwich ELISA Kit is a solid phase sandwich enzyme-linked immunosorbent assay (ELISA) that detects endogenous levels of PIP4K2C protein in a reduced assay time of 1.5 hours. Incubation of cell lysates and detection antibody on the coated microwell plate forms a sandwich with PIP4K2C in a single step. The plate is then extensively washed and TMB reagent is added for signal development. The magnitude of absorbance for the developed color is proportional to the quantity of PIP4K2C protein. Learn more about your ELISA kit options here.

*Antibodies in this kit are custom formulations specific to kit.

    Specificity / Sensitivity

The PathScan® RP PIP4K2C Sandwich ELISA Kit detects endogenous levels of PIP4K2C protein. The kit sensitivity is shown in Figure 1. This kit detects proteins from the indicated species, as determined through in-house testing, but may also detect homologous proteins from other species.

    Background

Phosphatidylinositol-5-phosphate 4-kinases (PIP4Ks) synthesize phosphatidylinositol-4,5-bisphosphate (PtdIns(4,5)P2), a key precursor in phosphoinositide signaling that directly modulates the activity of signaling proteins and cellular processes. There are two subfamilies of PIP kinases, type I and II, that generate PtdIns(4,5)P2 from distinct substrate pools. PIP4 type I kinases use PtdIns5P as a substrate, whereas PIP5 type II kinases use PtdIns4P (1,2). In mammalian cells, three isoforms of each PIP4K and PIP5K subfamily, encoded by distinct genes, have been characterized (3-7). All PIP kinases are stimulated by phosphatidic acid, extensively regulated by ARF and Rho GTPases, and inhibited by protein kinase A and PI-stimulated autophosphorylation (8).
PIP4K2C, also known as phosphatidylinositol-5-phosphate 4-kinase type-2 gamma (PI5P4Kγ), is a member of the type II PIP kinase family, and is broadly expressed with the highest levels in kidney and brain tissue (9). PIP4K2C can form a homodimer or heterodimer with its two other known family members, PIP4K2A and PIP4K2B (10). PIP4K2C is a regulator of mammalian target of rapamycin complex 1 (mTORC1) signaling, and Pip4k2c KO mice demonstrate enhanced immune responses and develop autoimmunity (11,12).
