    Product Usage Information

    Use as a blocking reagent to evaluate the specificity of antibody reactivity in western immunoblotting and dot blot protocols. For western blotting, add an equal volume of peptide as volume of antibody used in 10 ml total volume, and incubate at room temperature for 30 minutes before allowing to react with the blot. Recommended antibody dilutions can be found on the Bcl-xL Antibody #2762 and Bcl-xL (54H6) Rabbit mAb #2764 datasheets.

    Storage

    Supplied in 20 mM potassium phosphate (pH 7.0), 50 mM NaCl, 0.1 mM EDTA, 1 mg/ml BSA and 5% glycerol. 1% DMSO. Store at –20°C.

    Product Description

    This peptide is used to block Bcl-xL (54H6) Rabbit mAb #2764 and Bcl-xL Antibody #2762 reactivity.

    Quality Control

    The quality of the peptide was evaluated by reversed-phase HPLC and by mass spectrometry. The peptide blocks Bcl-xL (54H6) Rabbit mAb #2764 and Bcl-xL Antibody #2762 signal in both dot blot and western blotting.

    Background

    Bcl-xL prevents apoptosis through two different mechanisms: heterodimerization with an apoptotic protein inhibits its apoptotic effect (1,2) and formation of mitochondrial outer membrane pores help maintain a normal membrane state under stressful conditions (3). Bcl-xL is phosphorylated by JNK following treatment with microtubule-damaging agents, such as paclitaxel, vinblastine, and nocodazole (4,5).
