    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Leu140 of human DCPS protein.

    Background

    DCPS is a scavenger pyrophosphate and a member of the histidine triad family, which breaks down the mRNA cap structure during the decay process. During 3’-5’ mRNA decay, DCPS hydrolyzes m(7)GTP to generate m7GMP (1,2). DCPS also binds m(7)GDP strongly, which inhibits its activity against m(7)GTP (3). DCPS can shuttle from the nucleus to the cytoplasm and also plays a role in pre-mRNA splicing by affecting the nuclear cap-binding protein CBP20 (4). Additionally, DCPS can contribute to miRNA turnover, a function that is independent of decaying activity and dependent on exonuclease XRN2 (5). mRNA cap analogs that are resistant to DCPS activity have been widely utilized in the study of mRNA decay and stability (6-8). Inhibitors of decapping enzymes have been shown to be effective in spinal muscular atrophy mouse models (9).
