    Background

    Dynamin is a family of large GTPases that has been implicated in the formation of vesicles of both endocytotic and secretory processes (1). Dynamin plays an important role in the internalization of cell surface receptors, a process that attenuates the response to extracellular signals. There are three isoforms of dynamin. Dynamin 1 and 3 expression is enriched in the brain, while dynamin 2 expression is ubiquitous. All three isoforms mediate membrane fission during clathrin-mediated endocytosis. Dynamin 2 is also suggested to be involved in non-clathrin-mediated endocytosis and cytoskeletal interactions (2-4). Dynamin 3 is suggested to regulate AMPA (α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid) receptor recycling through its interaction with the postsynaptic density scaffolding proteins Homer and Shank (5-7). In addition to the role they play in neurotransmission, dynamin family members are considered therapeutic targets in cancer. Dysregulation of endocytosis in cancer leads to dysfunction in cell signaling, survival, and proliferation. Dynamin 1 and 2 have been shown to promote tumorigenesis, while dynamin 3 is a suggested tumor suppressor (8-12).
