Background
Estrogen receptor β (ERβ) is part of the nuclear receptor superfamily related to estrogen receptor α (ERα) and encoded by the ESR2 gene on chromosome 14 (1,2). Much like ERα, ERβ contains two activator functions, one in the N-terminal domain and one in the ligand binding domain. Both proteins have highly homologous DNA-binding domains and can synergistically regulate transcription (2,3). ERβ is tightly regulated by various mechanisms including estrogen binding, post-translational modifications, alternative splicing, circadian controls, and DNA methylation of the gene (4,5). The role of ERβ in various cancer types is controversial, but it is generally thought to be a tumor suppressor. Absence of ERβ has been shown to be a marker of poor prognosis in breast cancer and is correlated with tamoxifen resistance (6,7). In prostate cancer, ERβ can induce apoptosis, but its expression seems to be lost in higher grade tumors (8,9).
