    Product Usage Information

    For optimal ChIP results, use 2.5 μL of antibody and 10 μg of chromatin (approximately 4 × 106 cells) per IP. This antibody has been validated using SimpleChIP® Enzymatic Chromatin IP Kits.

    The CUT&RUN dilution was determined using CUT&RUN Assay Kit #86652.

    Monoclonal antibody is produced by immunizing animals with a recombinant protein fragment specific to human ETV4 protein.

    Background

    ETS translocation variant 4 (ETV4, PEA3, E1AF) is a member of the PEA3 (polyomavirus enhancer activator 3) subfamily of the E26 transformation-specific (ETS) DNA-binding transcription factors, which commonly bind a core DNA sequence of 5'-GGA(A/T)-3' (1,2). The PEA3 subfamily of ETV1, ETV4, and ETV5 are considered oncogenic transcription factors due to their overexpression and promotion of tumorigenesis in numerous cancers (2,3). ETV4 can be activated via the RAS-RAF-MEK-ERK pathway (2,4) or the PI3K/Akt pathway (5), and loss-of-function mutations of PEA3-family repressors such as CIC can increase ETV4 expression independent of either pathway (2,6). Though rare, 5' chromosomal fusions to ETV4 are drivers of metastasis in prostate cancer (7) and Ewing sarcoma (8). ETV4 regulates the expression of genes that control several developmental processes, including cell growth, differentiation, proliferation, survival, and cell cycle progression (9-11).
