    Background

    The mammalian ceramide synthases (CerS), also known as longevity assurance homologues (LASS), are a family of proteins that catalyze the formation of ceramide from acyl-CoA and sphingosine as part of the ceramide salvage pathway (1). Sphingosine derived from the lysosomal breakdown of complex glycosphingolipids can be delivered to the endoplasmic reticulum for LASS-dependent ceramide production (1). LASS family members exhibit preferences for different acyl-CoA fatty acid chain lengths (2-6) and have distinct tissue expression patterns, with notable enrichment of LASS1 in brain, LASS2 in liver and kidney, LASS3 in testis and skin, and LASS6 in intestine (6,7). Similarly, LASS proteins may function as tumor suppressors or promoters depending on tumor origin and metabolic context (8-11).
