    Background

    PAP-associated domain-containing protein 5 (PAPD5) and PAPD7, also known as TENT4B and TENT4A, respectively, are poly(A) polymerases involved in a layer of mRNA regulation by depositing guanosine residues on the poly(A) tail. This process, called mixed tailing, interferes with the deadenylase CCR4-NOT complex from efficiently deadenylating the poly(A) tail (1). PAPD5 and PAPD7 form a complex with ZCCHC14 and play a role in stabilization of hepatitis RNA (2-4). Small molecule inhibitors of PAPD5 and PAPD7 are being pursued for treatment against hepatitis B by inhibiting their polyadenylase activity (5,6).
