    Product Usage Information

    This product is the carrier free version of product #20302. All data were generated using the same antibody clone in the standard formulation which contains BSA and glycerol.

    This formulation is ideal for use with technologies requiring specialized or custom antibody labeling, including fluorophores, metals, lanthanides, and oligonucleotides. It is not recommended for ChIP, ChIP-seq, CUT&RUN or CUT&Tag assays. If you require a carrier free formulation for chromatin profiling, please contact us. Optimal dilutions/concentrations should be determined by the end user.

    BSA and Azide Free antibodies are quality control tested by size exclusion chromatography (SEC) to determine antibody integrity.

    Formulation

    Supplied in 1X PBS (10 mM Na2HPO4, 3 mM KCl, 2 mM KH2PO4, and 140 mM NaCl (pH 7.8)). BSA and Azide Free.

    Storage

    Store at -20°C. This product will freeze at -20°C so it is recommended to aliquot into single-use vials to avoid multiple freeze/thaw cycles. A slight precipitate may be present and can be dissolved by gently vortexing. This will not interfere with antibody performance.

    Specificity / Sensitivity

    Periostin (E5F2S) Rabbit mAb (BSA and Azide Free) recognizes endogenous levels of total human periostin protein. The antibody also recognizes an unknown weak signal around 40 kDa.

    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Ser395 of human periostin protein.

    Background

    Periostin (POSTN, OSF2) is a secretory matricellular protein that belongs to the fasciclin family. The protein has multiple functional domains that interact with different extracellular matrix (ECM) components (1-2). The N-terminal EMI domain of periostin interacts with fibronectin as well as collagens and lysyl oxidase. The middle part of the protein consists of four FAS 1 (fasciclin) domains which are responsible for the protein association with tenascin-C and CCN3. The C-terminal CTD domain assumes an inhibitory effect on FAS1 domain function. Lastly, the C-terminal HBD domain mediates proteoglycan association (1-2). Periostin has been shown to mediate important cellular processes such as ECM stemness, mechanical stress response, collagen cross-linking and fibrogenesis, cell proliferation, survival, and adhesion (1-3). It is an important modulator in multiple disease developments and has been frequently used as a biomarker for cancer, fibrosis, inflammation, and tissue wound healing (4-7).
