    Protocol

    Specificity / Sensitivity

Phospho-Parkin (Ser65) Antibody recognizes transfected levels of Parkin protein only when phosphorylated at Ser65.

    Species Reactivity:

    Human

    Source / Purification

Polyclonal antibodies are produced by immunizing animals with a synthetic phosphopeptide corresponding to residues surrounding Ser65 of human Parkin protein. Antibodies are purified by peptide affinity chromatography.

    Background

Parkin is a protein of 465 amino acids with an amino-terminal ubiquitin domain and a carboxy-terminal RING-box (1). In the case of autosomal recessive juvenile Parkinsonism (AR-JP), deletions have been found in the gene on chromosome 6 encoding the protein Parkin (2).
Parkin is phosphorylated by PINK1 at site Ser65. Phosphorylation at this site has been shown to be necessary for Parkin activation in vivo (3).
