    Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human pyruvate carboxylase protein.

    Background

    Pyruvate carboxylase (PC) catalyzes the carboxylation of pyruvate to oxaloacetate in an anaplerotic pathway to replenish TCA cycle intermediates. It is also critical in regulating gluconeogenesis in the liver (1). Studies show that the increase of PC flux contributes to the induction of hepatic gluconeogenesis by glucagon (2). In addition, high-fat diet is shown to promote hepatic glucose uptake and induce PC activity in the liver (3). Loss of hepatic PC prevents diet-induced hyperglycemia and insulin resistance. Furthermore, this enzyme is also required for antioxidant defenses (1).
