    Background

    Rad51 paralog D (Rad51D) is one of five Rad51 paralogs that form two distinct complexes, BCDX2 (Rad51B, Rad51C, Rad51D, and XRCC2) and CX3 (Rad51C and XRCC3) (1). Along with Rad51, these complexes are crucial in the repair of DNA double-strand breaks through homologous recombination (2), and in the response to DNA replication stress (3,4). Germline mutations in homologous recombination-associated genes, including those encoding Rad51C and Rad51D, lead to increased susceptibility to breast, ovarian, prostate, and pancreatic cancer (5).
