    Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, and 50% glycerol. Store at –20°C. Do not aliquot the antibody.

    Specificity / Sensitivity

    VGAT Antibody recognizes endogenous levels of total VGAT protein. Non-specific labeling in the crypts of small intestine is observed by immunofluorescence in fixed frozen mouse tissue. By western blot, this antibody detects a band of unknown origin at approximately 40 kDa.

    Polyclonal antibodies are produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human VGAT protein. Antibodies are purified by protein A and peptide affinity chromatography.

    Background

    The vesicular GABA transporter (VGAT), also known as the vesicular inhibitory amino acid transporter (VIAAT), loads GABA and glycine from the neuronal cytoplasm into synaptic vesicles for vesicular storage and subsequent exocytosis of these inhibitory amino acids. VGAT is present in both GABAergic and glycinergic neurons, with its expression limited to the brain (1,2). It can also recognize β-alanine as a substrate (2).

    Recently, tau accumulation, as found in Alzheimer’s disease (AD) patients, has been shown to selectively suppress the expression of the intracellular VGAT (3). At the same time, VGAT activation can help with anxiety exacerbated by AD pathology (4).
